Evercyte adds telomerized perinatal cells to immortalized range

Evercyte has added a range of telomerized perinatal cells that can be grown without limitations in vitro while maintaining cell type-specific markers and functions to its portfolio of immortalized cell lines.

These newly developed mesenchymal stromal cells (MSCs) are derived from different regions of the placenta and epithelial cells and provide valuable and standardizable in vitro models for studies such as those into the barrier function of the placenta or to perform toxicity assays.

Perinatal cell range

The four new additions are:

  • P-EP/SVTERT344 – Amniotic Epithelial Cells
  • P-MSC/TERT308 – Placental Amnion MSCs
  • CP-MSC/TERT308 – Placental Chorionic Plate MSCs
  • RA-MSC/TERT308 – Reflected Amnion MSCs

As with Evercyte’s other telomerized cell lines, these human perinatal cells and MSCs can be grown without limitations while maintaining expression of cell type-specific markers and function.

MSC-derived perinatal cells can be used to produce clinical-grade EVs. The cells were established under xeno-free conditions, with full documentation of any manipulation step and therefore can be transferred to a GMP facility to generate a master cell bank that can be used to produce clinical-grade extracellular vesicles.

Telomerized cell advantages

Primary cells maintain a phenotype like the corresponding cells in vivo. However, their application is limited due to their finite life span. Tumor cells, on the other hand, can be grown indefinitely but lose essential characteristics. These disadvantages of normal and tumor cell lines lead to the drawback that most in vitro bioassays cannot be reproduced and standardized.

To circumvent these limitations, Evercyte has focused on the establishment of telomerized cell lines that can be grown without growth arrest while maintaining a phenotype that resembles that of the corresponding primary/normal cells.

This is achieved by ectopic expression of the catalytic subunit of human telomerase and/or cell cycle regulators in primary cells followed by a detailed characterization of the cellular phenotype.
